Regulation of Th2 cytokine locus by Th2 locus control region
Th2 cytokine locus is coordinately regulated by enhancers, a silencer and a locus control region (LCR). Th2 LCR has been shown to contain several DNase I hypersensitive sites that are induced in Th2 cells, and to have a critical role in the intrachromosomal interaction. To examine the in vivo function of Th2 LCR, we generated CD4-specific Th2 LCR deficient (cLCR KO) mice using Cre-LoxP recombination. cLCR KO mice showed a dramatic reduction of Th2 cytokine expression. Deletion of Th2 LCR caused more closed chromatin conformation measured by H3 acetylation, H3-K4 methylation and DNA methylation in Th2 cells. Upon ovalbumin challenge in the mouse model of allergic asthma, cLCR KO mice showed marked reduction in airway inflammation and airway hyperresponsiveness. These results demonstrate that Th2 LCR is critically important in the regulation of Th2 cytokine genes, chromatin remodeling of Th2 cytokine locus, and in the pathogenesis of allergic asthma.
